Latent transforming growth factor beta binding protein (LTBP1) is composed of mature TGF-beta 1 dimer; latency associated peptide, and a latent TGF-beta binding protein (LTBP).
LTBP1 is a key regulator of TGF-β.
A whole CDS that encodes human LTBP1(L), is inserted in exon 2 of mouse Ltbp1 gene. The whole LTBP1(L) protein expression will be driven by endogenous mouse Ltbp1-L promoter, while mouse Ltbp1-L gene transcription and translation will be disrupted.
LTBP1(L) protein was detected in liver, lung, kidney, uterus and ovary of wild-type C57BL/6 mice and homozygous B-hLTBP1(L) mice, as the antibody can cross-recognize both human and mouse LTBP1(L).
Application: This product is used for pharmacodynamics and safety evaluation of drugs.
mRNA expression analysis
Strain specific analysis of LTBP1(L) mRNA expression in wild-type C57BL/6 mice and homozygous B-hLTBP1(L) mice by RT-PCR. Liver RNA was isolated from wildtype C57BL/6 mice (+/+) and homozygous B-hLTBP1(L) mice (H/H), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse or human LTBP1 primers. Mouse Ltbp1 mRNA was detectable only in wild-type C57BL/6 mice. Human LTBP1 mRNA was detectable only in homozygous B-hLTBP1(L) mice but not in wild-type mice.
Protein expression analysis
Strain specific LTBP1(L) expression analysis in homozygous B-hLTBP1(L) mice by western blot. Heart, liver, lung, kidney, uterus and ovary were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hLTBP1(L) mice (H/H), and analyzed by western blot with anti-LTBP1 antibody (Proteintech, 26855-1-AP). m/hLTBP1 was detectable in liver, lung, kidney, uterus, and ovary of wild-type C57BL/6 mice (+/+) and homozygous B-hLTBP1(L) mice.
