Orexin, a neuropeptide synthesized by hypothalamic neurons, exists in two distinct forms: orexin A and orexin B. These two types of orexin exert their physiological effects by binding to two specific G-protein-coupled receptors, namely orexin receptor 1 and orexin receptor 2. Any abnormalities in the functions of orexin and its associated receptors can disrupt the body's normal sleep-wake regulatory mechanism.
Gene targeting strategy for B-OX1R KO mice. The exons 1-7 of mouse Ox1r gene that encode the full-length protein were knocked out in B-OX1R KO mice.
mRNA expression analysis: Mouse Ox1r mRNA was detectable only in wild-type C57BL/6 mice but not in homozygous B-OX1R KO mice.
Application: This product is used for pharmacodynamics evaluation of Narcolepsy.

mRNA expression analysis
Strain specific analysis of Ox1r mRNA expression in wild-type C57BL/6 mice and B-OX1R KO mice by RT-PCR. Brain RNA were isolated from wild-type C57BL/6 mice (+/+) and homozygous B-OX1R KO mice (-/-), then cDNA libraries were synthesized by reverse transcription, followed by PCR with mouse Ox1r primers. Mouse Ox1r mRNA was detectable only in wild-type C57BL/6 mice but not in homozygous B-OX1R KO mice.
